What are pharmaceutical intermediates?

So-called pharmaceutical intermediates are, in fact, certain chemical raw materials or chemical products used in the synthesis processes of pharmaceuticals. These chemical products do not require a pharmaceutical manufacturing license and can be produced at ordinary chemical plants—as long as they meet certain quality standards, they can be used in pharmaceutical synthesis.
Increased demand
Over nearly 50 years of development, China has established a complete production system for β-lactam antibiotics. By 2012, China was able to produce almost all types of β-lactam antibiotics at very low costs. Penicillin production ranked among the highest in the world, and large quantities were exported to supply the international market. As for cephalosporin antibiotics, China is basically self-sufficient and even manages to secure a share for export.
In 2012, China was fully capable of producing all intermediates required for β-lactam drugs. With the exception of the parent nuclei 7-ACA and 7-ADCA—used in semi-synthetic drugs—which still required some imports—every side-chain intermediate could be produced domestically and was exported in large quantities.
Taking β-lactam antibiotics’ key intermediate, phenylacetic acid, as an example, China currently has nearly 30 manufacturers of phenylacetic acid, with a total annual production capacity of approximately 20,000 tons. However, most of these enterprises are relatively small in scale; the largest among them has an annual output of 2,000 tons, while most others produce only several hundred tons per year. In 2003, the country’s total domestic demand for phenylacetic acid was about 14,000 tons, with the following consumption breakdown: penicillin G accounted for 85%, other pharmaceuticals for 4%, fragrances for 7%, and pesticides and other applications for 4%.
With the development of China’s industries such as fragrances, pharmaceuticals, and agrochemicals, the demand for phenylacetic acid is set to increase further. By 2005, it is estimated that China’s pharmaceutical industry will consume approximately 14,000 tons of phenylacetic acid, the agrochemical industry will use 500 tons, and the fragrance industry will consume about 2,000 tons. Adding in consumption from other sectors, the total domestic demand for phenylacetic acid in 2005 is expected to reach 18,000 tons.
